While an undergraduate at the University of Chicago in the Hyde Park neighborhood of Chicago, Henner originated the role of "Marty" in the Kingston Mines production of Grease in 1971. She was offered the opportunity to reprise the role when the show was moved to Broadway. But she decided to take on the role of "Marty," in the National touring company, along with John Travolta who was playing "Doody". Henner is also credited with Broadway credits including Over Here! with Travolta and the revivals of Pal Joey. Social Security was also revived. The Tale of the Allergist's Wife is another Broadway credits. Her first film appearance in 1977's sleeper-hit Between the Lines. It was a comedy that featured then unknown actors John Heard and Lindsay Crouse. Her second role was opposite Richard Gere in the 1978 film Bloodbrothers.Henner gained notoriety for her role as Elaine Nardo in the situation comedy Taxi as an unmarried mother who works as a taxi driver who dreams to be an artist. She was the leading lady in the film of 1982 Hammett directed by Wim Wenders, which was produced by Francis Ford Coppola and starring her first husband Frederic Forrest. In 1983, Henner starred opposite Burt Reynolds in The Man Who Loved Women, produced by Blake Edwards. Reynolds then asked Henner to join the cast of Cannonball Run II later that year along with Shirley MacLaine and Dom DeLuise. She was the main character in the 1984 film Johnny Dangerously, playing the lover of Michael Keaton. In 1985 she once again played alongside John Travolta in Perfect. She was a co-star with Steve Martin as Trudi in L.A. Story in 1991. The performance earned her an American Comedy Award nomination as the Funniest Supporting Female Character in the field of Motion Picture. She was a co-star with Burt Reynolds from 1990 to 1994 in Evening Shade, a situation comedy that also featured Ossie Davis, Hal Holbrook, and Hal Holbrook. She was also seen in the film Noises Off (1992) as well as Man on the Moon (1999), which is a film that tells the story of Andy Kaufman, her Taxi co-star.
